Purification of Two Taxanes from Taxus cuspidata by Preparative High-Performance Liquid Chromatography

In the present study, an effective method of preparative high-performance liquid chromatography (Prep-HPLC) was established to purify two taxanes in Taxus cuspidata. During experimental operation, effects flow rate, injection volume, and column temperature on purity 10-deacetyltaxol (10-DAT) paclitaxel (PTX) were investigated, optimized conditions as follows: rate 10 mL/min, volume 0.5 mL, 30 °C. Under these conditions, 10-DAT PTX reached 95.33% 99.15%, respectively. The purified products characterized by scanning electron microscopy (SEM), (HPLC), electrospray ionization-high resolution mass spectrometry (ESI-HRMS). results demonstrated that HPLC can effectively from cuspidata with a >95%, which suitable for large-scale preparation PTX.
